Ok I had the bike running finally it would start fine with the choke on let it warm then it would idle but some time need a little throttle to keep it running
Now today it wouldn't rev up with the choke on would just sit at idle speed I tried adjusting the screws so it would idle without the choke on then it stopped
Now it won't even attempt to start without the choke on and atleast half throttle
Any ideas what the issue may be?
 
It could have a blocked pilot jet ?
What sort of bike and what carby is on it?

Turn the idle mixture screw right in (don't do it tight)
Now turn it back out till it's about 1.5 full turn's out.

Start the bike and let it idle and warm up.
Once it's warm you can adjust the mixture, turn the idle speed screw in about 1/2 a turn to raise the idle speed
Now adjust the mixture screw in or out 1/2 turn at a time, let it settle for 10 second's and listen to the way it run's.
You want to find the highest/smoothest point , so it may take a couple of tire's to get it right.
Once you think it's close to the best smooth idle, then you and turn the idle speed screw back out 1/2 or so till the idle is a normal speed.

Try the bike out and see how it goes,
if you need to adjust the mixtures now, just turn the mixture screw in a little 1/8 turn in or out 1/8 turn at a time to find the best setting, it won't be too far from the first time you adjusted it, and remember to let it settle idling for 10 second's or so after adjusting it.

If adjusting the idle mixture screw doesn't help then the fuel bowl need's to come off and the pilot jet will need to be cleaned, both the hole through it and the hole's across it.
Also check the hole at the base of the thread's where the pilot jet screw's into is clear, and that you can see light into the main bore of the carby.
